Abstract

Polygenic background modifies penetrance in hereditary cancer. We built a polygenic risk model in 18,000 carriers of high-penetrance variants.

Polygenic scores stratified lifetime risk by more than twofold within carriers, enabling personalised surveillance for hereditary cancer syndromes.
