Abstract

The tumour suppressor p53 safeguards genome integrity. We modelled p53 loss in organoids and tracked the emergence of structural variation.

p53 deficiency licensed chromothripsis and whole-genome doubling within ten passages, mechanistically linking tumour-suppressor loss to the genomic instability of carcinogenesis.
